Exploring high-quality whole-genome sequencing of FFPE samples from a large cancer cohort

For decades, tissue samples from resections and biopsies have been preserved by pathologists in formalin-fixed and paraffin-embedded (FFPE) blocks, resulting in sizeable tissue repositories that may be utilized for large-scale retrospective studies. Because these tissues are often paired with a great deal of clinical and outcome data, analysis of these archives has the potential to lead to major advances in cancer treatment and prevention.

Recent efforts have focused on utilizing next-generation sequencing to provide novel insights into different cancers, but this method has been limited by the poor quality of FFPE-derived DNA, sequencing libraries, and the sequence data. Good sequencing results could be obtained by limiting analyses to panels of select genes or whole-exome sequencing, but results from whole-genome sequencing (WGS) have remained poor.

Genuity Science (formerly known as WuXi NextCODE) has developed a process for generating high-quality WGS data using FFPE samples.  In this webinar, we will present the results from a large cancer sequencing project where this method was used to generate high-quality WGS data from FFPE tumor-normal sample pairs. An analysis of the sequencing yield, uniformity, and sequence and variant quality measures will be presented.
